过去40年过继细胞治疗的发展催生了多种新策略,例如嵌合抗原受体(CAR):将免疫细胞进行工程化改造后输注给患者,以清除癌细胞。在此背景下,CAR-T 细胞治疗血液系统恶性肿瘤已显示出显著前景。然而,实体瘤和血液系统恶性肿瘤中均存在多重障碍,限制CAR-T 疗效。因此,近年来CAR-NK和CAR-M细胞疗法作为新选择出现,旨在应对CAR-T 治疗相关挑战。目前,世界各地正在开展多种CAR免疫细胞疗法的人体临床试验,目标是增强抗肿瘤活性并降低毒性。本文全面综述CAR免疫细胞疗法用于多种人类恶性肿瘤的近期研究,并讨论过去几年中出现的挑战。

Advancements in adoptive cell therapy over the last four decades have revealed various new therapeutic strategies, such as chimeric antigen receptors (CARs), which are dedicated immune cells that are engineered and administered to eliminate cancer cells. In this context, CAR T-cells have shown significant promise in the treatment of hematological malignancies.

However, many obstacles limit the efficacy of CAR T-cell therapy in both solid tumors and hematological malignancies. Consequently, CAR-NK and CAR-M cell therapies have recently emerged as novel therapeutic options for addressing the challenges associated with CAR T-cell therapies.

Currently, many CAR immune cell trials are underway in various human malignancies around the world to improve antitumor activity and reduce the toxicity of CAR immune cell therapy. This review will describe the comprehensive literature of recent findings on CAR immune cell therapy in a wide range of human malignancies, as well as the challenges that have emerged in recent years.
