过去十年CAR-T 疗法的进步推动多种癌症治疗发生变革。尽管已取得显著成功,复杂制备流程和治疗相关毒性等障碍仍妨碍其广泛应用。作为替代方案,嵌合抗原受体工程化NK 细胞(CAR-NK)疗法提供了一种可行机会,可形成更简单、成本效益更高的“现货型”治疗方式,且可能减少不良反应。本综述共纳入71项研究。研究者检索并回顾了PubMed、Web of Science和Scopus数据库中的符合条件研究。基于文献数据,综述认为CAR-NK细胞疗效会因肿瘤模型、剂量、CAR代际和扩增方法等因素而显著变化。综述还考察了不同构建体及不同代CAR-NK细胞对抗肿瘤活性的影响。

Advancements in the field of CAR-T therapy have brought about a revolution in the treatment of numerous types of cancer in the past ten years.

However, despite the remarkable success achieved thus far, certain barriers impede the widespread implementation of this therapy such as intricate manufacturing processes and treatment-associated toxicities. As an alternative, chimeric antigen receptor-engineered natural killer cell (CAR-NK) therapy presents a viable opportunity for a simpler and more cost-effective "off-the-shelf" treatment option, which is likely to result in fewer adverse reactions.

A total of 71 studies were included in this review. Eligible studies were searched and reviewed from the databases of PubMed, Web of Science and Scopus. Based on data extracted from articles, we concluded that CAR-NK cell efficiency can vary considerably depending on factors such as tumor model, dosage, CAR generation and expansion method.

Furthermore, investigating consequences of utilizing various constructs and generations of CAR-NK cells on their anti-tumor activity examined in this review.
