嵌合抗原受体(CAR)细胞疗法在患者治疗中的重要性日益提高。CAR-T 细胞已被证明可有效治疗血液系统恶性肿瘤,但该疗法仍存在潜在移植物抗宿主病(GVHD)、神经毒性和细胞因子释放综合征(CRS)等治疗相关风险。因此,CAR NK 细胞疗法有望成为新的治疗选择。NK 细胞是细胞毒性淋巴细胞,可通过精确识别并清除恶性细胞,支持机体针对自身免疫性疾病(AD)和癌细胞的先天免疫反应。对 NK 细胞进行基因修饰,可通过 CAR 依赖和非依赖机制,为 AD 和癌症治疗提供双重策略。CAR 细胞疗法已成功用于癌症治疗,促使研究者进一步探索这一创新疗法在 AD 等其他疾病中的应用。CAR-T 与 CAR NK 细胞的互补作用推动了该领域的发展。本文综述这些细胞治疗癌症和 AD 的最新疗效研究。

Chimeric Antigen Receptor (CAR) based therapies are becoming increasingly important in treating patients. CAR-T cells have been shown to be highly effective in the treatment of hematological malignancies.

However, harmful therapeutic barriers have been identified, such as the potential for graft-versus-host disease (GVHD), neurotoxicity, and cytokine release syndrome (CRS). As a result, CAR NK-cell therapy is expected to be a new therapeutic option. NK cells act as cytotoxic lymphocytes, supporting the innate immune response against autoimmune diseases and cancer cells by precisely detecting and eliminating malignant cells.

Genetic modification of these cells provides a dual approach to the treatment of AD and cancer. It can be used through both CAR-independent and CAR-dependent mechanisms. The use of CAR-based cell therapies has been successful in treating cancer patients, leading to further investigation of this innovative treatment for alternative diseases, including AD. The complementary roles of CAR T and CAR NK cells have stimulated exploration in this area.

Our study examines the latest research on the therapeutic effectiveness of these cells in treating both cancer and ADs.
